Immunoassay: Principles, Types, and Applications in Modern Science

Immunoassay is a widely used biochemical technique designed to detect and measure specific substances such as proteins, hormones, or drugs in a sample by using the principle of antigen–antibody interaction. It plays a crucial role in clinical diagnostics, pharmaceutical research, environmental monitoring, and food safety testing. Due to its high sensitivity and specificity, immunoassay has become one of the most reliable analytical methods in modern laboratories.

What is Immunoassay?

Immunoassay is based on the ability of antibodies to bind selectively to specific antigens. When a target molecule (antigen) is present in a sample, it interacts with a corresponding antibody, forming an antigen–antibody complex. This reaction can be detected and measured using various labeling techniques such as enzymes, fluorescence, or radioactivity, allowing precise quantification of the target substance.

Types of Immunoassay

There are several types of immunoassay techniques, each serving different diagnostic and research purposes:

Enzyme-Linked Immunosorbent Assay (ELISA): One of the most common methods used for detecting antigens or antibodies in blood samples.

Radioimmunoassay (RIA): Uses radioactive isotopes to measure concentrations of specific antigens, known for high sensitivity.

Fluorescent Immunoassay (FIA): Utilizes fluorescent markers for detection and is commonly used in automated systems.

Chemiluminescent Immunoassay (CLIA): Relies on light emission from chemical reactions, offering high accuracy and speed.

Applications of Immunoassay

Immunoassay techniques are widely applied across various fields. In clinical diagnostics, they help detect diseases such as HIV, hepatitis, and hormonal disorders. In pharmacology, they are used to monitor drug levels and therapeutic responses. Environmental scientists use immunoassays to detect pollutants and toxins in water and soil, while the food industry relies on them for identifying contaminants and allergens.

Advantages of Immunoassay

One of the key advantages of immunoassay is its high sensitivity, allowing detection of even trace amounts of substances. It is also highly specific due to the unique antigen–antibody interaction. Additionally, immunoassays are adaptable for automation, making them suitable for high-throughput testing in clinical laboratories.
